DNA Test Accuracy in New Zealand: Understanding Accuracy vs Probability of Paternity

DNA testing is widely regarded as the gold standard for determining biological relationships. However, two important concepts are often misunderstood: accuracy of the test and probability of paternity. While both are essential, they measure very different things. Understanding the difference helps ensure you have confidence in both the testing process and the results.

What Is DNA Test Accuracy?

The accuracy of DNA testing refers to how reliably and correctly a laboratory analyses genetic markers to determine whether two individuals share a biological relationship. Accuracy reflects the quality of the laboratory processes, not the outcome of the test.

Factors affecting accuracy include:

  • Laboratory processes: Accredited laboratories that comply with standards such as ISO/IEC 17025 follow strict quality systems to ensure results are highly reliable.

  • Technology: Modern genetic analysers and validated test kits reduce the chance of error.

  • Sample quality: Poor-quality samples (e.g. degraded or contaminated DNA) can impact testing, though experienced labs have procedures to manage this.

Accuracy in numbers: In a well-run laboratory, accuracy is extremely close to 100%, meaning the likelihood of a technical error influencing your result is exceptionally low.

Probability of Paternity

The probability of paternity (POP) is a statistical measure of how likely it is that a tested man is the biological father of a child, compared to an unrelated random man in the population.

It is calculated after the DNA profiles are analysed and is expressed as a percentage. Typically, results are either:

  • Zero (0%) for exclusions (not the biological father), or

  • Extremely high (e.g. 99.9999%) for inclusions (very likely to be the biological father).

How Is Probability of Paternity Calculated?

  • The shared genetic markers between the child and the alleged father.

  • The frequency of those markers in the wider population (population genetics data).

  • The “prior odds” of paternity – usually set at 50:50, assuming equal chances that the man is or is not the father before the DNA evidence is considered.

Why it’s not “accuracy”: Probability of paternity is not about the reliability of the lab test. Instead, it reflects the statistical strength of the biological relationship. Even a perfectly accurate test can produce a POP of 0% (not the father) or 99.99% (almost certainly the father).
